When one room lags behind

Why duct runs break down
Flexible duct flattens when it is forced through a tight bay or used as a step during attic work. Metal runs drift apart at their crimped ends as framing shifts, especially when the hangers were sparse to begin with. Cloth tape that sealed the seams on day one dries out within years and lets go. The thermostat never shows any of it. What you notice is one room that stays a few degrees behind.
When a section has to be replaced
Some sections are past saving, flex that has been crushed, metal that has rusted through or a run that was too small from the outset. In those cases we fit new duct matched to the size of what it joins, seal each joint with mastic and hang it so it does not lean on other ducts. That almost never means a full new system. Usually it is one run, replaced properly.
